Sedentary phytoparasitic nematodes are capable to maneuver the cell cycle in addition to disarray the cytoskeleton in their favor. Our projects focus on the investigation of cell cycle progression and the cytoskeleton organization and dynamics in feeding sites induced by parasitic nematodes. To study the cytoskeletal changes and cell cycle in root cells undergoing feeding cell development we make use of immunocytochemical approaches, in vivo GFP strategies, histological analysis and infection tests to investigate the undergoing changes in both processes within feeding sites at different stages of nematode infection. Functional analysis of lines down-regulated and knockout, or overexpression of genes involved in the cytoskeleton and the cell cycle have been carried out as well. Recently we deciphered the function of g-tubulins as nucleating new microtubule cytoskeleton in organizing centers in feeding sites. We have also identified genes such as CCS52 gene family, DEL1 and RHL1 involved in the endocycle control of gall cell. Moreover, we newly demonstrated that cell cycle inhibitor genes of the ICK/KRP family negatively control gall size and nematode reproduction. A potential link between cell cycle and the cytoskeleton is also an approach that will be pursued.
